Our laboratory is interested in the molecular, preclinical and clinical pharmacology of nonsteroidal androgens and anticancer agents. In 1998, we were the first to report the discovery of nonsteroidal androgens (i.e., small molecules that mimic the effects of testosterone). We are now examining the structure and function of the human androgen receptor, identifying novel and important structure-activity relationships for binding and activation of the receptor, and characterizing the in vivo pharmacology, pharmacokinetics, and metabolism of these drugs using animal models of relevant diseases. Current projects include: (1) molecular and preclinical pharmacology of irreversible antiandrogens, (2) examination of the molecular endocrinology, pharmacokinetics, pharmacology, and metabolism of nonsteroidal androgens, and (3) studies of novel thiazolidines and bis-indoles as potential drugs for the treatment of prostate cancer.. These projects are currently funded by the National Institutes of Health and the Department of Defense, U.S. Army Prostate Cancer Research Program. Additional studies are focused on the effects of these drugs on coactivator recruitment by the androgen receptor and potential therapeutic applications of these drugs, including osteoporosis, hypogonadism, and male contraception.
